Genetix have announced a new on-line guide introducing an automated methodology that significantly accelerates the production of high titre monoclonal antibodies and protein biologicals using mammalian cells in cultures.

Located at www.genetix.com, the guide illustrates how you can drastically reduce the time taken to produce high yields hybridomas and adherent cells compared to conventional techniques. The entire process, from post fusion selection, growing cells in petri dishes, visualising, picking and dispensing clones into microwells, through to incubation may be automated and improved using the system as shown.

Use of semi-solid media combined with on-board imaging on the automated ClonePix system, which is more sensitive and has higher resolution than the human eye, means that individual clones can be picked earlier compared to manual methods. Furthermore, the novel properties of the ClonaCell semi-solid media (StemCell Technologies Inc.*) media ensure each colony is monoclonal, therefore eliminating the need for rounds of cell sorting and sub-cloning. The operator can change picking parameters such as intensity, colony size, circularity and distance apart to ensure clonality. Each colony can be tracked from dish to microwell, and the parameters and images stored or exported. This enables the investigator to refine parameters and develop protocols to maximise production methodologies. Data shows that the proportion of ELISA positive clones is experimentally the same for colonies picked with ClonePix as for those picked manually. From a financial perspective, estimates indicate the payback period for investment in ClonePix is short (data available on request) and break-even is under 2,000 colonies per week.
