INTEGRA Biosciences (www.integra-biosciences.com) has announced a new application note describing the use of the VACUSAFE comfort for optimising DNA purification based upon the widely used 'bind-wash-elute' procedure. This procedure involves releasing the nucleic acids from the cells, binding the DNA onto a matrix (usually packed in a column), washing impurities off the matrix-bound DNA and finally eluting the DNA.

In the described experiment plasmid DNA (pBluescript) from an overnight culture of transformed DH5a E. coli was extracted with a commonly used kit. To investigate the effect of different levels of applied vacuum on the effectiveness of the DNA purification, the cell lysate containing the DNA was drawn through a matrix-packed column using a range of applied vacuum. The results of the experiment clearly showed that the DNA extraction kit performed best at lowest vacuum settings probably because this allowed maximal binding of DNA onto the column matrix.

The compact, benchtop VACUSAFE comfort is the system of choice for almost any application requiring a controlled vacuum. Incorporating the latest pump technology the VACUSAFE comfort offered the infinitely variable, gentle vacuum control enabling optimal DNA purification procedure results.

The VACUSAFE comfort is extremely simple to operate - simply switch on and set the required vacuum. As soon as the target vacuum has been reached, the pump switches off automatically. If the vacuum collapses in use, the pump switches back on automatically.

The VACUSAFE comfort incorporates all the essential elements for safe and productive removal of virtually any liquid. Waste is collected in a shatterproof 3-litre glass or 4-litre polypropylene container that is fitted with an overflow filter and optionally a level sensor to prevent liquids from getting into the pump. All parts of the VACUSAFE comfort that may come in contact with liquid may be repeatably sterilised, guaranteeing maximum safety even with hazardous and critical liquids.
